- Does the introduction explain the objective of the research presented in the preprint?
- Yes
- Are the methods well-suited for this research?
- Highly appropriate
- Are the conclusions supported by the data?
- Somewhat supported
- Reporting gaps exist in several places, which I have already marked, and informed the authors in an email.
- Are the data presentations, including visualizations, well-suited to represent the data?
- Somewhat appropriate and clear
- Some representations like the genomic sequence are missing. The discrepancy in the fluoroscence colours in the immunohistochemistry images are not properly clarified. Accession numbers of Secondary and primary antibodies used have also not been clearly mentioned. I have detailed all te discrepancies in my email to te authors.
- How clearly do the authors discuss, explain, and interpret their findings and potential next steps for the research?
- Very clearly
- Is the preprint likely to advance academic knowledge?
- Highly likely
- Would it benefit from language editing?
- Yes
- I have already added my comments regarding the possible edits in a copy and sent it over to the authors for their feedback.
- Would you recommend this preprint to others?
- Yes, but it needs to be improved
- The preprint is an interesting read. It talks about a potential alternate pathway for regulating the myelination of oligodendrocytes and demethylation of the DNA in MS, which will probably lead to a suitable treatment for the disease in future. Statistical methods used need revision and elaboration, cohort studies and sample sizes are unclear for some experiments and some data have reporting gaps. Also, the language is confusing at places. I have already emailed the authors with the detailed review of what needs to be addressed further to make this paper publication-ready.
- Is it ready for attention from an editor, publisher or broader audience?
- Yes, after minor changes

Use of Artificial Intelligence (AI)
The author declares that they used generative AI to come up with new ideas for their review.
1. Cross-checked the statistical methods used. 2. Refined the clarity of the language in the author queries and grouped similar queries together. 3. Cross-referenced the preprint with the papers in the reference list. 4. Double -checked whether the in-text citations and the reference list were aligned and whether any in-text citations were missing.
