Age-Related Diminution of the LINC Complex Component SUN2 in Human Mesenchymal Stromal Cells

Identifying reliable molecular markers for biological aging is essential for under-standing age-related cellular dysfunction and developing targeted therapeutic inter-ventions. This study investigated the relationship between chronological age andexpression of ENSG00000100242 (SUN2) in human mesenchymal stromal/stem cells(MSCs). Analysis of gene expression data revealed a significant negative correlationbetween SUN2 expression and donor age (r =−0.451, p = 0.00027). Mean ex-pression was 5.31 ± 0.31 (range: 4.60− 5.90). Linear regression analysis yielded anintercept of 5.77 and a slope of−0.0082, confirming the inverse relationship. SUN2encodes a component of the linker of nucleoskeleton and cytoskeleton (LINC) com-plex, which maintains nuclear integrity and regulates gene expression. The observedage-dependent downregulation of SUN2 suggests its potential role as a biomarkerfor cellular aging and its contribution to age-related functional decline in MSCs.
